Characterization of Newcastle Disease Virus Isolated from Peacocks in Palembang City, South Sumatra

Introduction: Newcastle Disease (ND) is an infectious disease in various types of poultry caused by the Virus (NDV). Cases ND Indonesia have been reported commercial and backyard chickens, pigeons, ducks geese, even eagles peacocks. Peacock a wild bird protected Indonesia's Government Regulation No. 7 1999. This study aims to identify, isolate characterize NDV molecularly cases diagnosed as Method: Samples were obtained from organs (lungs spleen) peacocks which showed neurological symptoms, diarrhoea then died. Real-time RT-PCR was used identify cause death peacock. isolation observation embryonic changes carried out on chicken eggs. Sequencing F HN entire genes NDV. The nucleotide sequences analyzed using MEGA-X software, including amino acid prediction, analysis genetic variation at level, homology construction phylogenic tree. Result: results sample identification positive for virus. Observations embryos are stunted, few feathers, haemorrhagic, die less than 60 hours. with titer 26. Molecular that RRQKRF cleavage site pattern gene had 95.8-97.6% same branching area previous virus Indonesia. There no mutations antigenic site, glycolysis neutralization epitopes gene. Conclusion: isolated peacock velogenic strain NDV, subgenotype VII.2 has close range previously domestic poultry. result shows also threat birds.
